Afraxis is discovering and developing drugs to treat diseases through the modulation of p21-activated kinase (PAK). Afraxis’ initial indication of interest is Fragile X syndrome with future expansion into other CNS diseases, including schizophrenia and autism spectrum disorders. By inhibiting PAK, a protein that regulates the development and maturation of synaptic connections that are defective in mental retardation syndromes and other CNS diseases, Afraxis will target CNS disease modification rather than just treat symptoms. PAK is also implicated in a number of different oncological settings and Afraxis is exploring the potential of PAK inhibitors for the treatment of cancers such as neurofibromatosis, breast cancer and lung cancer.
Ambit Biosciences, Inc. is engaged in the discovery and development of small-molecule kinase inhibitors for the treatment of cancer.
Ambit’s lead product candidate, AC220, is an oral, small molecule FMS-like tyrosine kinase-3 (FLT3) inhibitor being developed for the treatment of Acute Myeloid Leukemia (AML), the most common type of blood cancer in adults. FLT3 is mutated and activated in 25-40% of AML patients, causing poor prognosis and decreased response to existing treatments including chemotherapy and stem cell treatments. AC220 has entered a Phase 2 pivotal trial after obtaining concurrence from the FDA for its co-primary endpoint as a single, open-label trial which may be eligible to support registration (marketing approval), depending on the results it will generate. Called the ACE trial (AC220 monotherapy Efficacy study), this clinical trial will enroll a total of 180 patients who have refractory or relapsed AML with a mutated FLT3 kinase called internal tandem duplication (FLT3-ITD), which has negative prognostic implications.
Ambit’s second clinical candidate, AC480, is a pan-HER (human epidermal growth factor receptor) small molecule tyrosine kinase inhibitor; HER kinases (e.g., HER-1 or EGFR, HER-2, HER-3, HER-4) and the attendant signaling transduction pathway have been found to correlate with, and contribute to, a variety of human malignancies, including breast, gastric, lung, head and neck, and colorectal cancer. AC480, licensed from Bristol-Myers Squibb, has shown unique and promising results in pre-clinical animal cancer and human xenograft model (e.g., synergistic effects with paclitaxel, another widely-used chemotherapeutic drug), including the ability to cross the blood-brain barrier, a characteristic that would be useful in the treatment of brain tumors and metastases.
Founded in 2005, AnaptysBio, Inc. is a privately-held company focused on the generation of antibody therapeutics, and the leader in the use of somatic hypermutation (SHM) for antibody discovery and optimization. SHM is the body’s natural process for generating potent antibodies. AnaptysBio’s proprietary SHM-XEL platform, which couples fully human antibody libraries with in vitro somatic hypermutation (SHM) in mammalian cells to generate high affinity lead candidates, replicates key features of the human immune system and overcomes limitations of prior antibody technologies. By harnessing the natural mechanism of antibody maturation under controlled conditions, SHM-XEL allows for the selection of optimal antibody properties such as high affinity, functionality, cross-reactivity and epitope diversity. AnaptysBio has established broad intellectual property around the use of SHM and is currently building a pipeline of novel therapeutic antibody product candidates. The Company has signed partnerships with Merck, Roche, Novartis, Celgene, Gilead and DARPA. Major investors in AnaptysBio include founding investor Avalon Ventures, as well as Alloy Ventures, Frazier Healthcare Ventures, and Novo A/S.
Aratana Therapeutics, Inc., based in Kansas City, is a companion-animal health company, leveraging human pharmaceutical and biotechnology drug development to identify compounds that can be developed into novel therapeutics for dogs and cats. Companion animal health provides numerous market advantages over human health. Animal health drug development poses unique regulatory challenges, but with the experienced Aratana drug development team, and their ability to pick the right compounds at the right stage, new medicines can be developed in significant less time and in a very cost efficient manner, compared to human drug development. Another significant advantage is that in veterinary medicine, there are no third party payer issues, and pet owners are eager to purchase state of the art medicines for their pets, who are increasingly viewed as family members. In addition, there are 10 multi-billion-dollar companies that provide therapeutics to companion animals and fewer than a dozen biotechs servicing this group (compared to 20 big pharmas and 2000+ biotechs). The focus is on developing drugs for major diseases and unmet needs for companion animals, using a capital efficient drug development model to streamline the path to regulatory approval. By identifying molecules with specific attributes, compounds can be developed in a cost-effective and expedited timeline.

Syndax Pharmaceuticals is a late-stage clinical development oncology company with a platform based on an epigenetic approach to overcoming the problem of resistance in cancer therapy. Syndax holds worldwide rights to entinostat, an oral, highly selective, class I histone deacetylase (HDAC) inhibitor, which inhibits cancer-relevant HDAC enzymes that contribute to epigenetic alterations driving cancer growth and drug tolerance. Targeted therapies represent an important class of agents used to treat both breast and lung cancers based on blocking the tumor’s addiction to growth promoting signals. Over time these therapies become ineffective due to acquired resistance. There are more than 300,000 individuals living with incurable breast or lung cancer in the United States. Entinostat’s unique pharmacokinetic properties, convenient oral dosing and HDAC selectivity, maximize the potential to safely combine with and extend the benefit of proven cancer targeted therapies.
